摘要
目的:探讨三七总甙抗肝纤维化作用机制。方法:按常规方法分离正常大鼠肝星状细胞,用三七总甙药物温育液作用于传一代培养的肝星状细胞,~3H-TdR掺入观察细胞增殖,ELISA法测定上清及细胞层Ⅰ型胶原含量,改良Lowry's法测定细胞层总蛋白以校正细胞数。结果:三七总甙不同浓度组均能抑制肝星状细胞增殖及细胞内外Ⅰ型胶原生成,尤以细胞外的抑制更为明显,与对照组及2.5×10^(-7)M秋水仙碱组比较差异显著。结论:三七总甙抗肝纤维化作用可能是通过抑制肝星状细胞增殖及细胞内外Ⅰ型胶原生成来实现的。
Aim: To research the mechanism of antihepato fibrosis by the main glycocide of Panax pseudo ginseng. Method: The hepato astrocytes from normal ratd following routine, then put the cells subcultured in the solution of the main glycocide. Later, observed the proliferation of the cells by 3H-TdR, tested the content of collegen I in the supernatant and cell's layer by using the ELISA method and tested the content of total protein by using the improved Lowry's method. Result: All of the various concentration of the main glycocide could suppress the proliferation of hepato astro-cyte and the production of collagen I . The difference compared with the control group and the Colchicine (2.5×10-7 M ) group were significant. Conclusion: It may be through the suppression of the hepato-astrocyte's proliferation and the collagen I 's production to repress the hepato fibrosis by the main hlycocide.
