摘要
目的:为探讨降钙素基因相关肽(CGRP)对高血压病(EH)患者动脉血管平滑肌细胞(VSMC)的一氧化氮合酶(NOS)和一氧化氮(NO)产生的作用。方法:对EH患者和血压正常者(NT)的动脉VSMC进行培养,分别测定EH组和NT组VSMC的NO含量和NOS活性,观察EH患者VSMCNO的产生以及CGRP对其的影响。结果:①基础状态下,EH组VSMC的NO含量和NOS活性分别为42.73±6.76μmol/(2.5×106cels)和0.24±0.05nmol/(2.5×106cels),均显著低于NT组的74.52±4.37μmol/(2.5×106cels)和0.55±0.10nmol/(2.5×106cels)(P<0.05);②在CGRP作用下,EH组的NO含量和NOS活性分别为88.63±4.12μmol/(2.5×106cels)和10.55±1.28nmol/(2.5×106cels),均显著低于NT组的100.49±9.69μmol/(2.5×106cels)和12.45±1.61nmol/(2.5×106cels)(P<0.05);③基础状态和CGRP作用下两组VSMC的NO含量和NOS活性随?
Aim: To investigate the effects of calcitonin gene related peptide(CGRP) on the
nitric oxide synthase (NOS) activity and nitric oxide(NO)production of vascular smooth muscle
cells (VSMCs) in essential hypertensive(EH)patients Methods: The VSMCs of EH patients and
the normotensives (NT) were cultured We measured the NO contents and NOS activities of
the VSMC in the EH and NT groups separately, observed the NO production of the VSMCs in the
EH patients and the effect of CGRP on it Results:Under basal conditions, the NO content and
NOS activity of the VSMCs in the EH group were 4273676 mol/25106 cells and 024005
nmol/25106cells respectively, which were significantly lower than 7452 437 mol/25106 cells
and 055010 nmol/25106cells of the NT group(P<005); (2)With the effect of CGRP, the NO content
and NOS activity of the EH group were 8863 412 mol/25106 cells and 1055128 nmol/25106cells
respectively, which were remarkably lower than 10049969 mol/25106 cells and 1245 161
nmol/25106celnitric oxide nitric oxide synthase vascular smooth muscle cells ls (P<005) of
the NT group;Under basal conditions and with the effect of CGRP, the NO contents and NOS
activities of the two groups significantly increased with the prolongation of culture
time(P<005);(4)With the effect of CGRP, the NO content and NOS activities of both groups
increased and enhanced significantly as compared with those under basal conditions(P<001)
Conclusion: The NOS activity and NO content reduced in VSMC of the EH patients,and CGRP
could enhance the activity of NOS and increase the content of NO, suggesting that the
deficiency of CGRP might be involved in the pathogenesis of EH
