STEAP4蛋白在人脂肪组织中表达大小的验证

Verification on Expression Size of STEAP4 Protein in Human Adipose Tissue

目的采用STEAP4多克隆抗体对其在人脂肪组织中的表达大小进行验证。方法 6例阑尾炎手术患儿中随机选取3例患儿的脂肪组织标本,抽提总蛋白,应用本实验室自制的STEAPG蛋白多克隆抗体,采用Westernblot技术检测STEAP4蛋白在人脂肪组织中表达的大小。结果目的条带出现于蛋白Marker相对分子质量55000以下位置,与STEAP4蛋白预测相对分子质量52000基本相符。结论本实验室自制的STEAP4蛋白多克隆抗体具有良好的免疫学活性,能够满足STEAP4免疫印迹检测的实验要求。STEAP4蛋白在人脂肪组织中的相对分子质量约为52000,为后续深入研究STEAP4蛋白的功能奠定了基础。

Objective To verify the expression size of STEAP4 protein in human adipose tissue.Methods Three adipose tissue was randomly selected from 6 paients with appendicitis who had been operated,and the total protein was extracted.The prepared polyclonal antibody against STEAP4 was used.Western blot was performed to verify the expression size of STEAP4 protein in human adipose tissue.Results The result of Western blot showed that there was only a band below 55 000 approximately equates with the predicted molecular weight of STEAP4 protein（52 000）.Conclusions The prepared polyclonal antibody against STEAP4 has favourable immunologic competence,and STEAP4 protein revealed 52 000 in human adipose tissue,which will be helpful for further study of STEAP4.
