三七花总皂苷抑制人乳腺癌细胞侵袭的体外研究 被引量:8

Inhibitory Effects of Panax Notoginseng Flower Saponins on Invasion of Human Breast Cancer Cell in Vitro

摘要 目的:研究三七花总皂苷对人乳腺癌细胞株MDA-MB-231侵袭能力的影响并探讨其相关机制。方法:MTT实验检测三七花总皂苷对肿瘤细胞增殖的抑制作用;Transwell小室法进行肿瘤细胞侵袭实验;RT-PCR检测β1-integrin、MMP-2、EGFR mRNA表达的变化;Western blotting检测β1-integrin、EGFR蛋白表达的变化。结果:在浓度为50~200μg/ml时,三七花总皂苷对人乳腺癌细胞的增殖具一定的抑制作用。Transwell小室实验显示,三七花总皂苷在浓度为10、50、100μg/ml均表现出侵袭抑制作用,浓度为50μg/ml侵袭抑制效果最佳,达到32.0%。三七花总皂苷对各浓度组对MDA-MB-231细胞β1-integrin、MMP-2和EGFR基因表达有下调作用(P<0.01),一些浓度组对β1-integrin和EGFR蛋白的表达也具下调作用。结论:三七花总皂苷通过下调β1-integrin、EGFR mRNA和蛋白的表达,抑制肿瘤细胞与ECM粘附,降低肿瘤细胞运动能力,从而抑制人乳腺癌细胞的侵袭转移。 Objective:To study the effects of Panax notoginseng flower saponins on invasion of human breast cancer cell line MDA-MB-231 and explore the possible mechanisms. Methods: The inhibitory effects of Panax notoginseng flower saponins on cancer cell proliferation were detected by MTT; the ability of invasion of cancer cell was evaluated by transwell assay; the expressions of β1-integrin,MMP-2 and EGFR mRNA and the expressions of β1-integrin、EGFR protein were tested by RT-PCR and western blotting respectively. Results: Panax notoginseng flower saponins inhibit the proliferation of human breast cancer cell at the concentration of 50 to 200 μg/ml. Transwell assay showed that Panax notoginseng flower saponins manifested inhibitory effects on the proliferation of human breast cancer cell at the concentrations of 10,50,100 μg/ml and the inhibitory effects were the best at the concentration of 50μg/ml reaching to 32.0%. The gene expressions of β1-integrin,MMP-2 and EGFR in MDA-MB-231 cell were decreased treated by Panax notoginseng flower saponins at all kinds of concentrations and the protein expressions of β1-integrin and EGFR were also decreased at some concentrations. Conclusion: Panax notoginseng flower saponins prevent the adhering of tumor cell and ECM,decrease the mobility of tumor cell and inhibit the invasion and metastasis of human breast cancer cell through depressing mRNA and protein expressions of β1-integrin and EGFR.
