摘要
目的在于观察体外培养条件下,库普弗细胞(KC)贮脂细胞(FSC)对肝细胞(PC)基质生成调节效应及PC增殖及合成功能的影响。采用FSC、KC和PC的分离培养,观察KC和FSC对PC增殖活性和DNA含量的影响,应用免疫细胞化学和图像分析系统对PC中Ⅰ、Ⅱ、Ⅳ型胶原和纤维粘连蛋白(FN)进行定量分析,以及用液闪法检测PC中3H-脯氨酸掺入量。结果证明FSCncm可明显促进PCDNA合成,PC可合成Ⅰ、Ⅲ、Ⅳ型胶原及FN。FSCncm可促使双核的PC增多,线粒体分裂。提示肝细胞可合成Ⅰ、Ⅲ、Ⅳ型胶原及FN;PC内可能直接组装胶原纤维;KCncm、KCtcm、FSCtcm均可上调PC的基质合成能力。
J We observed that effect of Kupffer Cells(KC) and Fat Store Cells(FSC) to liver parenchymal cells(PC) matrix svnthesis regulating and PC proliferation and function. We adopted separation cultured of FSCKC and PC,observed effect of KC and FSC to proliferation activity and DNA content of PC. It used immunocytochemistry and image pattern analysis system to quantitative analysis collagen type and fibnectin(FN) in PC,and applied liquid scintillation method to detecting incorporation number of 3Hprolin in PC.FSCncm can promote obviously PC DNA synthesis.PC can synthesis collagen type and FN.FSCncm may impel two nucleus PC increasing and mitochondria cleavage.The hepatocytes can synthesis collagen type and FN,It may directly compose collagen fiber in PC,All of KCncmKCtcmFSCtcm can promote PC synthesis ability of matrix.
