白藜芦醇对酒精性肝损伤大鼠血清中TNF-α、IL-6、ICAM-1水平的影响及机制 被引量:7

Effects of resveratrol on serum TNF-α,IL-6,ICAM-1 in alcohol-induced hepatic injury in rats

摘要 目的:研究白藜芦醇(resveratrol,Res)对酒精性肝损伤后炎性因子水平的影响和NF-κB信号通路的变化。方法:大鼠灌胃给予55度红星二锅头复制肝损伤模型。随机分为正常对照组、肝损伤模型对照组、Res低剂量组(25mg/kg)、Res中剂量组(50mg/kg)、Res高剂量组(100mg/kg)、联苯双酯组成(100mg/kg),每日给药2次,连续7d。以Elisa试剂盒分别测定血清TNF-α、IL-6、ICAM-1含量和肝组织NF-κBp65、磷酸化IκB-α水平。结果:与正常对照组相比,酒精性肝损伤模型大鼠血清中TNF-α、IL-6、ICAM-1水平和肝组织NF-κB、磷酸化IκB-α水平显著升高。与模型对照组相比,Res显著降低血清TNF-α、IL-6、ICAM-1含量,抑制肝组织NF-κB、磷酸化IκB-α水平。结论:Res通过抑制NF-κB、降低炎性因子水平,对于酒精性肝损伤的肝功能具有保护作用。 Objective:To investigate the effect of resveratrol(Res) on inflammatory cytokines and NF-κB pathway.Methods:The rats were randomly divided into 6 groups,including the normal control group,alcohol-induced hapatic injuried model control group,Res 25 mg/kg group,Res 50 mg/kg group and Res 100 mg/kg group and bifendate group 100 mg/kg.The hepatic injuried model was imitated by po 55° Red Star Erguotou.Res and bifendate were administrated twice daily,continuously for 7 days.The concentrations of serum TNF-α,IL-6,ICAM-1 and the contents of liver NF-κB p65,phosphorylating IκB-α were determined by the commercial Elisa kits.Results:Compared with the normal control group,the concentrations of serum TNF-α,IL-6,ICAM-1 and the contents of liver NF-κB p65 in liver were significantly increased in the model group(P〈0.01).Res significantly reduced the concentrations of serum TNF-α,IL-6,ICAM-1 and the contents of liver NFκB p65,phosphorylating IκB-α compared with the model group(P〈0.01).Conclusion:Res provented liver from alcoholic impairment through decreasing cytokine release followed supressing supressing NF-κB p65,phosphorylating IκB-α expressing.
