摘要
目的:研究白刺总黄酮及单体化合物对血管内皮细胞损伤的保护及修复。方法:用高糖(HG)诱导血管内皮细胞损伤,用酶标仪检测细胞内一氧化氮(NO)、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)以评价氧化的程度。结果:不同浓度的白刺总黄酮及单体化合物槲皮素、山奈酚、异鼠李素可促进高糖受损的内皮细胞修复,同时提高SOD、NOS、GSH-PX活力和NO水平。结论:白刺总黄酮具有保护血管内皮细胞损伤的作用,与其单体化合物槲皮素、异鼠李素相比,作用效果较强,与山奈酚作用效果相当,其机制与抗氧化作用有关。
Objective:To evaluate the effects of compound of flavonoid and flavonoids of Nitraria tangutorun Bobr.on the injured vascular endothelial cells and its mechanism.Methods:Human umbilical vascular endothelial cells(HUVEC) viability from high glucose injury was assessedby nitrogen monoxidum(NO),nitric oxide synthase(NOS),superoxide dismutase(SOD),glutathione peroxidase(GSH-PX) in the cell culture fluid with model SN-3001 Microplate Spectrophotometer.Results:Flavonoids of different concentration increased the repair of impaired HUVEC and increased the ratio of SOD、NOS、GSH-PX activity and NO.Conclusion:These results demonstrate that quercetin,campherol,isorhamnetin can produce the protective action on high glucose injured cultured HUVEC.Compare flavonoids with quercetin,isorhamnetin,the flavonoids is better and same to campherol.Its mechanism of action may be related to antioxidant effect.
