痹祺胶囊对骨关节炎的抗氧化作用

The antioxidation and protective effects of Biqi Capsule on the experimental osteoarthritis

目的:通过观察兔骨关节炎模型血清、尿、组织匀浆中氧自由基相关生化指标,研究痹祺胶囊对骨关节炎软骨的抗氧化作用。方法:以Hulth法复制兔膝骨关节炎模型,解剖显微镜下观察软骨组织并予评分,以正常对照组、假手术对照组,检测各组兔血清和尿液的丙二醛(MDA)含量、血清和膝关节滑膜组织超氧化物歧化酶(SOD)活性及一氧化氮(NO)含量,并设痹祺胶囊低、中、高浓度组及西药组对照以了解其抗氧化保护作用。结果:解剖显微镜下观察各治疗组间关节软骨性状无差异,均优于模型对照组;痹祺胶囊中、高剂量组血清、尿MDA及NO含量低于模型对照组水平,SOD活性高于模型对照组水平,且呈一定的剂量依赖性。结论:痹祺胶囊具有对抗过氧化、保护关节软骨的作用。

Objective：To observe the antioxidation of Biqi Capsule（BQC） on the rabbit model osteoarthritis by determining the morphology scale with a dissecting microscope and the malonaldehyde（MDA） content,superoxide dismutase（SOD） activity and nitric oxide（NO） content in blood serum,urine and membrana synovialis of experimental rabbits.Methods：A rabbit model of knee joint osteoarthritis（OA） was established by anterior cruciate ligament transection,and antioxidation Biqi Capsule on the articular cartilage was evaluated.Animals were randomly divided into seven groups：the normal control group,the sham-operation group,the anterior cruciate ligament transaction group as model set,the BQC 0.168g/kg treated group,the BQC 0.336g/kg treated group,the BQC 0.672g/kg treated group,the Glucosamine Sulfate Capsules（GSC） 0.2g/kg treated group.The medial and lateral femoral condyle,and the medial and lateral tibial plateau of the knee joints were surveyed with a dissecting microscope and the grading of pathological changes of them were measured.The MDA content in blood serum and urine of rabbits was determined,the SOD activity and the NO content in blood serum and synovialis homogenate were detected.Results：There were no significant statistical differences between the pathological scale of each treated group.The MDA content in urine of 0.168g/kg and in blood serum and synovialis homogenate of 0.336g/kg and 0.672g/kg BQC treated rabbits,and the NO content in blood serum and synovialis homogenate of 0.336g/kg and 0.672g/kg BQC treated rabbits were significantly decreased compared with the model group（P〈0.05）.And the SOD activity in blood serum and synovialis homogenate of 0.336g/kg and 0.672g/kg BQC treated rabbits were significantly increased compared with the model group（P〈0.05）.Conclusion：Biqi Capsule can protect the arthrodial cartilage of rabbit model osteoarthritis by the antioxidation effects.
