摘要 目的观察心气虚证模型大鼠学习记忆和脑内皮层部位胆碱能系统的变化,探讨心气虚证模型大鼠学习记忆能力下降的机制,阐明中医"心主神明"的物质基础。方法 SD大鼠随机分为5组:正常对照组、疲劳对照组、小剂量普萘洛尔+疲劳组、中剂量普萘洛尔+疲劳组和大剂量普萘洛尔+疲劳组,采用强迫跑步加灌服普萘洛尔的方法建立心气虚证大鼠模型,采用Morris水迷宫法检测大鼠学习记忆能力,生化比色法检测乙酰胆碱酯酶(AchE),放免法检测M受体。结果大剂量普萘洛尔+疲劳组出现学习能力的降低,皮层AchE活性显著增高,M受体Rt值显著降低。结论心气虚证模型大鼠出现脑内胆碱能功能紊乱,可能是造成学习记忆能力下降的物质基础。 Objective To observe the changes of cortex cholinergic system in rats of heart-Qi deficiency syndrome,explore the possible mechanism of learning-memory deficit in rats of heart-Qi deficiency and illustrate one of traditional Chinese medical theory that the heart governs the mental activities. Methods SD rats were randomly divided into 5 groups:normal (N),exercise-induced fatigue (F),small dose propanolol + exercise-induced fatigue (SF),middle dose propanolol + exercise-induced fatigue (MF) and great dose propanolol + exercise-induced fatigue groups (LF). The rat models of heart-Qi deficiency syndrome were established by using complex factors including forced running till exhaust and gavage propanolol. The ability of learning and memory were measured by Morris water maze test,biochemical colorimetry was applied to determine the activity of acetylcholine esterase (AchE). M receptor was estimated by radio-immunity method. Results In LF group,ability of learning-memory descended,the activity of AchE in cortex increased and Rt value of M receptor decreased obviously. Conclusions The dysfunction of cholinergic system possibly is the material basis of learning-memory deficit in rats of heart-Qi deficiency.
