6种中药提取物对胃癌细胞株细胞毒作用及多西紫杉醇增敏作用的研究

The anti-proliferation and sensitization effect to docetaxel of six traditional Chinese medicine extracts on gastric cancer cell lines

目的中医药是我国特有的民族医药宝库,为抗癌药物的研发提供了丰富的资源。文中研究6种中药提取物对2株胃癌细胞系增殖抑制作用并挑选具有多西紫杉醇(Docetaxel,DOC)增敏作用的中药提取物。方法采用MTT法研究6种中药提取物(藤黄酸、白藜芦醇、连翘安博立酸提取物、连翘达玛烷提取物、汉防己甲素、蛇床子素)对于胃癌细胞SGC-7901、BGC-823细胞毒作用。观察低细胞毒剂量的中药提取物对DOC敏感性的影响。采用C I指数法判断连翘达玛烷提取物与DOC的协同作用。结果藤黄酸对于胃癌细胞株增殖抑制作用最强、汉防己甲素次之,白藜芦醇、蛇床子素及连翘提取物细胞毒作用较差。低剂量连翘达玛烷提取物在胃癌2株细胞株具有较强的DOC增敏作用。SGC-7901细胞中等抑制作用时连翘达玛烷提取物与DOC具有协同作用,BGC-823细胞中高抑制作用时2种药物具有显著的协同作用。结论 6种中药提取物具有一定抑制肿瘤细胞增殖的作用。其中,连翘达玛烷提取物具有较强的DOC增敏作用。

Objective Traditional Chinese Medicine（TCM） offers a rich source of anticancer reagents.In current study,we studied on the anti-proliferation and sensitization effects on docetaxel of six kinds of TCM extracts on gastric cancer cell lines.Methods The anti-proliferation effect of TCM extracts（gambogic acid,resveratrol,extracts of Fructus Forsythiae containing ambrolic acid or dammarane compounds,tetrandrine and osthol） on human gastric cancer cell lines SGC-7901 and BGC-823 were evaluated by using MTT assay.Low-doses of cytotoxicity were chosen to observe the potential sensitization effect on docetaxel.Combined index was calculated to assume the potential interaction between dammarane extract of Fructus Forsythiae（DA） as indicated previously.Results Gambogic acid presented the strongest anti-proliferation effect with tetrandrine ranked sencondly,while the other four extracts showed minor cytotoxicity.Low-dose ammarane extract of Fructus Forsythiae showed strong sensitization effect to docetaxel.When evaluated using CI method,DA and docetaxel showed synergistic effect on both SGC-7901cells and BGC-823 cells.Conclusion Six TCM extracts had potential anti-proliferation effects and dammarane extract of Fructus Forsythiae showed strong sensitization effect to docetaxel on SGC-7901 and BGC-823 cells.
