摘要
目的:评价缺氧/复氧后血管内皮细胞ICAM1表达及内皮白细胞粘附率的变化。方法:体外培养大鼠骨髓血窦内皮细胞,缺氧90min,分别给予复氧1h,2h,4h,8h,12h,24h,用半定量ELISA法测ICAM1的表达量,同时还观察内皮细胞对中性粒细胞的粘附率。在复氧时加入药物NAC、灯盏花,复氧后4h观察ICAM1的表达。结果:(1)体外培养的内皮细胞缺氧后ICAM1表达上调,随复氧时间的延长ICAM1呈增强趋势。(2)药物灯盏花、NAC对复氧4h的ICAM1表达无抑制。(3)内皮细胞对白细胞的粘附率在缺氧后增加,复氧使粘附更强。ICAM1抗体或CD11b抗体抑制复氧后的粘附增强,而灯盏花、NAC则无抑制作用。结论:(1)缺氧/复氧导致血管内皮细胞ICAM1表达上调,加强内皮白细胞粘附。(2)复氧时加入灯盏花、NAC不能抑制ICAM1表达。
Objective: To
evaluate the expression of ICAM1 and the adhesive rate of neutrophils with endothelial cells
after anoxia and reoxygenation. Method: The endothelial cells of rat bone marrow were cultured
in vitro.Endothelial cells monolayers were exposed to anoxia for 90 min,reoxygenate for 1h,2h,
4h,8h,12h,24h,expression of ICAM1 was assayed by ELISIA,and the adhesion between
endothelial cells and neutrophils were measured.The endothelial cells monolayers also were
treated with handmazz or NAC after reoxygenation. Results :(1)Increased expression of ICAM1
appeared in cultured endothelial cells after anoxia and enhanced after
reoxygenation.(2)Handmazz or NAC couldn't inhibit ICAM1 expressin after reoxygenation
4h.(3)The adhesive rate of neutrophils with endothelial cells increased by anoxia and
reoxygenation.AntiICAM1 antibody or CD11b antibody could suppress it,but handmazz or NAC
had no affect. Conclusion:5BZ(1)The increased expression of ICAM1 and adhesion between
endothelial cells and neutrophils were induced by anoxia/reoxygenation.(2)Treated with
handmazz or NAC after oxygenation couldn't inhibit ICAM1 expression.
