不同产地连翘药材的高效液相色谱特征图谱研究 被引量:9

摘要 目的:建立连翘药材的高效液相色谱特征图谱分析方法。方法:采用SinoChrom ODS-BP(200mm×4.6mm,5μm)色谱柱,以乙腈(A)-水(B)为梯度流动相,线性梯度洗脱程序为:0~5min,2%~10%A;5~17min,10%~18%A;17~40min,18%~21%A;40~55min,21%~35%A;55~70min,35%~55%A。流速1.0mL·min-1,柱温25℃,检测波长231nm。结果:通过对11批不同产地的连翘药材的测定,标定10个共有峰,并通过"中药指纹图谱相似度评价软件",计算出9批药材相似度在0.9以上,2批药材相似度在0.9以下。结论:该方法准确可靠,重复性好,为更好地控制连翘药材的内在质量标准,提供了科学依据。 OBJECTIVE To establish an HPLC fingerprint method for the quality control of Forsythia suspensa (Thunb.) Vahl. METHODS The chromatographic fingerprints were determined on a SinoChrom ODS-BP coiunm (20 cm× 4. 6 mm,5 μm) with the gradient elution solvent system composed of acetonitrile (A) and water (B) . The elution program was 0 - 5 min, 2%- 10%A;5-17 min,10%- 18% A;17-40 min, 18%- 21% A;40-55 rain,21%-35% A;55-70 rain,35%- 55% A. The flow rate was 1.0 mL·min^-1, the colunm temperature was maintained at 25 ℃ and the wavelength was aequired at 231 nm. RESULTS 10 co-possessing peaks were selected as the fingerprint peaks of Forsythia suspensa (Thunb.) Vahl from 11 production places. The similarities,nine production places were proved to be higher than 0. 9 and two production places were proved to be less than 0.9, were caculated with the help of "The Similariy Caculation Soft of Fingerprint Chromatography of Traditonal Chinese medicine. "CONCLUSION This method with good characteristics and specificity can be used in the quality control of Forsythia suspensa (Thunb.)Vahl.
