摘要
目的:制备与分析抗可提取性核抗原(ENA)单克隆抗体;方法:以ENA为抗原,免疫BALB/c小鼠,取其脾细胞与小鼠Sp2/0骨髓瘤细胞融合,经反复筛选和再克隆化,建立能稳定分泌抗ENA单克隆抗体(McAbs)的杂交瘤细胞株;结果:共获得11株能稳定分泌抗ENA的杂交瘤细胞株。对其中的5种单克隆抗体(IG5,2E5,2C6,4C10和4E6)进行了初步分析,结果如下:小鼠腹水抗体效价范围2×10-5~3×10-6;免疫球蛋白亚类鉴定,1株为IgG2a,1株为IgG2b,其余3株均为IgG1;
