摘要
以廉价的商业化产品作为原材料,以提高发酵液中的青春双歧杆菌活菌数为目的,通过单因素试验筛选并最终确定能够用于规模化生产青春双歧杆菌的廉价工业化培养基成分,即:脱脂乳8%(W/V)、双歧因子A1%(W/V)、双歧因子B1%(W/V)。结果表明,在此培养基基础上,筛选出了青春双歧杆菌的最佳生长条件:最佳接种量为1%(V/V)、最佳培养温度为37℃。在此优化的生长条件下,测定青春双歧杆菌的生长曲线和pH值变化,选取在厌氧条件下培养时间9h为最佳培养时间,此时青春双歧杆菌活菌数最高可达1.16×109cfu·mL-1。
In this study,the low-priced commercialized products were screened forindustrial medium composition to enhance the number of the viable Bifidobacterium adolescentic.A single factor test was carried out to determine the industrial medium components for B.adolescentic.Finally,the optimum industrial medium consisted of 8%(W/V)skim milk,1%(W/V)bifidobacterium factor A and 1%(W/V) bifidobacterium factor B.Using the above optimized medium,the optimum growth condition was obtained two aspects:the inoculum amount was 1%(V/V)and growth temperature was 37 ℃.Under this optimum growth condition,the growth curve and the change of pH value were monitored.According to the growth curve,the optimum terminal time was chosen at 9 h under the anaerobic condition.On this time point,the No.of viable B.adolescentic was 1.16×109 cfu·mL^-1.
