摘要
目的观察五没食子酰葡萄糖对内毒素的体内拮抗作用。方法动态浊度法测定五没食子酰葡萄糖对小鼠内毒素血症的影响。观察大鼠内毒素攻击后,五没食子酰葡萄糖对外周血内毒素的中和作用及对肿瘤坏死因子-α释放的抑制作用。最后观察了五没食子酰葡萄糖对致死量内毒素攻击小鼠的保护效果。结果在小鼠体内五没食子酰葡萄糖具有较强的内毒素中和作用,两者之间具有明确的量效关系。五没食子酰葡萄糖可明显降低内毒素攻击大鼠血浆内毒素和肿瘤坏死因子-α水平,且对肿瘤坏死因子-α的抑制作用与内毒素变化密切相关。五没食子酰葡萄糖能够明显延迟动物的死亡时间,显著降低死亡率。结论五没食子酰葡萄糖可能通过对内毒素的结合及中和作用,在体内发挥拮抗内毒素作用。
Objective To study the effect of anti-endotoxin of β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose in vivo.Methods The endotoxin-neutralizing effect of β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose was assayed in endotoxemic NIH mice with LAL,and the neutralization of endotoxin and suppression of tumor necrosis factor-a release were also observed in lipopolysaccharide-challenged rats.We also tested whether β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose could protect mice from a lethal challenge by lipopolysaccharide.Results β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose significantly decresed the plasma endotoxin level in endotoxin mice and rats,which was closely related to the tumor necrosis factor-a level.Animals pretrated with β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose showed a delayed death time and were protected from lipopolysaccharide-induced lethality.Conclusion β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose develops the effect of anti-endotoxin in vivo,which is associated with the role of neutralizing endotoxin.
