摘要
目的观察宣肺定喘汤对哮喘大鼠气道重构和炎症介质的抑制作用。方法 6周龄清洁级SD雄性大鼠60只,随机分为空白对照组、宣肺定喘汤大剂量组、宣肺定喘汤中剂量组、宣肺定喘汤小剂量组、地塞米松组及病理模型组6组,建立大鼠哮喘模型,比较各组大鼠支气管和肺组织形态学的改变及肺组织中Eotaxin、ICAM-1表达水平;外周血液中炎性细胞、IgE表达水平;肺泡灌洗液中炎性细胞及Eotaxin、ICAM-1表达水平。结果与空白对照组相比,病理模型组支气管和肺组织形态学有显著的改变,炎性细胞、Eotaxin、ICAM-1、IgE的表达水平显著升高(均P<0.01)。与病理模型组相比,各治疗组炎性细胞、Eotaxin、ICAM-1、IgE的表达水平有显著差异(P<0.05,P<0.01)。光镜观察宣肺定喘汤大剂量组支气管及肺组织结构基本正常。结论宣肺定喘汤能有效抑制气道重构和炎症介质的释放,以达到平喘的目的 。
Objective Objective To explore Xuanfei Dingchuan Decoction on airway remodeling and inhibition of inflammatory mediators in asthmatic rats.Methods 60 clean grade male SD rats of 6 weeks old were randomized into blank control group,Xuanfei Dingchuan Decoction high-dose group,medium-dose group and low-dose group,dexamethasone group and pathological model group.Rats were induced Asthma and compared the morphological changes of bronchial and lung tissues,and the expression levels of Eotaxin,ICAM-1 of Lung tissue of each groups;the expression levels of inflammatory cells,IgE of peripheral blood;and,the expression levels of the inflammatory cells and Eotaxin,ICAM-1 in bronchoalveolar lavage fluid.Result Comparing with the blank control group,the bronchial and lung tissue morphology of pathological model group significantly changed,and the expression levels of inflammatory cells,Eotaxin,ICAM-1,IgE have significantly increased.Comparing with the pathological model,the expression levels of inflammatory cells,Eotaxin,ICAM-1,IgE in each treatment groups showed significant differences(P0.05)or extreme significant differences(P0.01).We found that the bronchial and lung tissue of Xuanfei Dingchuan Decoction high-dose group were almost normal by light microscopy.Conclusion Xuanfei Dingchuan Decoction can effectively inhibit airway remodeling and the releasing of inflammatory mediators to realize the objective of treating asthma.
