HPLC-ELSD Determination of Liquidambaric acid in Tongmai Ningxin Granules

目的建立测定通脉宁心颗粒中路路通酸含量的方法。方法样品经过无水乙醇超声处理后,采用HPLC-ELSD测定路路通酸的含量,色谱条件,采用Elite Kromasil C18色谱柱,以甲醇-0.25mol/L醋酸溶液(72∶28)为流动相,流速:1.0mL/min,柱温:30℃,漂移管温度45℃,载气为N2,流速3.5 mL/min。采用外标两点法计算含量。结果路路通酸在1.8-36.0μg/mL浓度范围内有良好线性关系(r=0.9998),平均加样回收率为99.09%,RSD为1.17%。结论本方法快速简便,结果准确,重复性好,可用于该制剂中路路通酸的定量分析。

Objective Objective To establish a method for the determination of liquidambaric acid in Tongmai Ningxin granules.Method The sample was extracted in EtOH by microwave.The liquidambaric acid was determined by HPLC-ELSD.Elite Kromasil C18 column was used,the mobile phase consisted of Methanol-0.25mol/L acetic acid（72∶28）,the flow rate was 1.0 mL/min,the column temperature was 30℃,the temperature of drift tube was 45℃,nitrogen gas as carrier gas,the flow rate was 3.5 mL/min,with a universal detector Evaporation Light Scattering Detector（ELSD）.Calculate contents with external standard two-point method.Results The linear response range of liquidambaric acid was 1.8-36.0μg/mL（r=0.9998）.The average recovery was 99.09%,RSD was 1.17%.Conclusions The method is simple,accurate and repeatable,it can be applied in quantitative determination of liquidambaric acid in Tongmai Ningxin granules.
