摘要
目的:探讨枸杞多糖在肝癌细胞血管内皮生长因子(VEGF)表达调控中的作用。方法:体外培养高转移性人肝癌细胞株HCCLM3,分为对照组、枸杞多糖浓度5μmol/L,10μmol/L,15μmol/L等4组,处理6小时后,通过MTT法检测枸杞多糖对肝癌细胞株HCCLM3增殖的抑制程度,采用Westernbolt技术观察VEGF蛋白表达的变化,而后用20μmol/L,处理6小时后,采用免疫荧光技术观察VEGF蛋白表达的变化。结果:枸杞多糖可抑制肝癌细胞的增值,使肝癌细胞HCCLM3的VEGF蛋白的表达下降,这种抑制作用随着枸杞多糖浓度增大而增大(P<0.01),呈正相关关系,用免疫荧光技术观察VEGF蛋白表达的变化时,发现当LBP浓度为20μmol/L时,VEGF蛋白的表达完全被抑制。结论:枸杞多糖对体外培养的人肝癌细胞(HCCLM3)的生长有明显抑制作用,其作用机制与VEGF有很大的关系。
Objective:To investigate the roles of Lycium barbarum polysaccharide (LBP) in the regulation of expression of vascular endothelial growth factor(VEGF) in hepatocellular carcinoma (HCC) cells.Methods:The high metastatic cell line HCCLM3 were cultured in DMEM medium.HCCLM3 cells were divided into control groups and LBP treatment groups (5 μmol/L,10 μmol/L,15 μmol/L),the cell proliferation was measured by MTT and the expression of VEGF protein was detected by Western bolt after 6 h.Results:In HCCLM3 cells,the cell proliferation was inhibited by LBP,the expression of VEGF protein can be apparently inhibited by LBP.With the increase of LBP concentration,the expression of VEGF protein decreased gradually(P0.01) and was in a dose dependent manner.Conclusion:LBP can reduce the expression of VEGF mRNA in HCCLM3 cell line,and then inhibit cell proliferation.
