系统性红斑狼疮患者外周血单个核细胞中Brn-3a mRNA表达水平的研究

摘要 目的 探讨系统性红斑狼疮(SLE)患者外周血单个核细胞中Brn-3a mRNA 表达及其与疾病活动性的关系.方法 以β-actin为内参照,建立逆转录实时荧光定量聚合酶链反应(FQ-RT-PCR)方法,在Light Cycler Real time PCR检测仪上检测92例SLE患者(活动期46例,缓解期46例)、其他结缔组织病(CTD)患者30例(疾病对照组)、健康体检者30例(正常对照组)外周血单个核细胞(PBMCs)中Brn-3a mRNA 的表达水平和含量.以ΔCt=Ct(待测基因)-Ct(内参基因)来比较基因表达水平的高低.结果 活动期SLE患者Brn-3a mRNA表达水平明显高于缓解期SLE患者(P〈0.01);活动期SLE患者Brn-3a mRNA表达水平明显高于正常对照组与疾病对照组(P〈0.01);缓解期SLE患者Brn-3a mRNA表达水平与正常人及疾病对照组差异无统计学显著性意义(P〉0.05).结论 活动期SLE患者Brn-3a mRNA表达水平显著高于正常人、疾病对照组和疾病缓解期,提示Brn-3a的表达与SLE的发生发展存在一定关联性,可能参与了SLE的发病过程并与疾病的活动性有关. Objective To evaluate the expression of Brn-3a mRNA on peripheral blood mononuclear cells (PBMCs) in patients with systemic lupus erythematosus(CSLE). To find out the relationship between Brn-3a and disease activity. Methods The expression of Brn-3a mRNA were detected by reverse transcriptlon-reat time quantitative polymerase chain reac- tion (FQ-RT-PCR) on PBMCs in 92 patients with SLE(46 active patients,46 remittent patients),30 patients with other rheumatic diseases as diseases controls,and 30 health controls. β-aclin were used as endogenous control. The results were calculated by the formula △Ct(objective gene)-Cl(endogenous reference gene). Results The values of Brn-3a mRNA expression in active patients were higher than that in remittent patients(P〈0.01),patients with other rheumatic discases (P 〈 0. 01) and healthy controls (P〈0. 01). There were no significant difference among remittent patlents,other rheumatic diseases patients and healthy controls(P〈0.05). Conclusion The study indicates that the levels of Brn-3a mRNA expression on PBMCs is significantly elevated in patients with active SLE,and it may be involved in pathogenesis of disease.
