枳菊解郁汤对抑郁模型小鼠学习记忆及脑内c-fos表达的影响

摘要 探讨枳菊解郁汤对慢性应激抑郁模型小鼠学习记忆的影响及其机制.将小鼠随机分为正常对照组,模型组,阳性药物组(氟西汀),枳菊解郁汤低剂量组、中剂量组和高剂量组.通过开场和Morris水迷宫实验,检测各组小鼠行为和学习记忆能力的变化;采用免疫组织化学方法检测c-fos在海马和前脑皮层的表达.结果显示,慢性应激抑郁模型组小鼠自发活动和探究行为明显降低(P<0.01);空间学习记忆能力明显下降(P<0.05);海马CA1、CA3和DG区及前脑皮层c-fos表达明显下降(P<0.05).与模型组小鼠相比,枳菊解郁汤中高剂量明显改善小鼠的行为和学习记忆功能,以及海马和前脑皮层c-fos的表达.结果提示,枳菊解郁汤的抗抑郁作用可能与海马和前脑皮层c-fos表达的上调有关. In this paper,we study the effects of Zhijujieyu-tang on the ability of spatial learning-memory and the expression of c-fos in chronic stress induced-depression model mice brain.Kunming mice were randomly divided into six groups:the normal control group,the model group,the positive control (fluoxetine) group,the high,medium and low concentration groups of Zhijujieyu-tang.The behaviors and the ability of spatial learning-memory of mice were determined by open-field test and Morris Water Maze,and the expression of c-fos in the brain were detected by immunohistochemical method.The results showed that the spontaneous activities and exploratory behaviors were remarkably decreased in the model group mice (P0.01),and the spatial learning and memory in them were significantly decreased (P0.05).The c-fos expression in the CA1,CA3,DG regions of hippocampus (HP) and prefrontal cortex (PFC) were significantly reduced in the model group mice (P0.05).Zhijujieyu-tang medium and high dosage groups can improve the behavior,the learning and memory function,and the expression of c-fos in HP and PFC.The results indicate that Zhijujieyu-tang possesses a certain antidepressant-like property and its mechanism may be closely related to the up-regulation of c-fos expression in HP and PFC.
