摘要
分别采用不同培养基(MS、1/2MS、GD和SH)、外植体(茎尖、幼嫩茎段、嫩叶和老叶)、0~100.00mg/LNAA和不同基因型木薯品种对木薯初生体细胞胚发生及植株再生的影响进行研究。结果表明,与其他3种培养基(1/2MS、GD、SH)相比,在MS培养基上,木薯品种GR911初生体细胞胚及植株再生的发生率最高,分别为36.0%和8.3株/外植体;以幼嫩茎段为外植体时,虽然GR911初生体细胞胚发生率及植株再生频率稍低于茎尖,但诱导效果较好且省时省工。随着MS培养基中NAA浓度的增加,GR911的初生体细胞胚发生率和植株再生频率呈先增加后降低趋势,以添加60.00mg/LNAA的诱导效果最好,可以直接诱导初生体细胞胚发生和芽的形成。此外,供试的9个木薯品种中有8个品种(GR911、SC5、SC6、SC7、SC8、SC9、南植188和SC205)可直接由茎段诱导初生体细胞胚发生和植株再生,但不同基因型木薯品种间的体细胞胚发生率和植株再生频率差异较大,以SC8最高,SC7最低。
The induction of primary somatic embryogenesis and plant regeneration in cassava were studied by using different culture media,viz.,MS,1/2MS,GD and SH;type of explants,viz.,shoot tip,young stem and leaf,old leaf;different NAA concentrations(0-100.00 mg/L) and different cassava cultivars.The results showed that the primary somatic embryogenesis and plant regeneration induction rate of cassava variety GR911 were higher in MS medium compared to other three basic culture media.When young stem was used as explant,although the primary somatic embryogenesis and plant regeneration induction rate in GR911 was lower than that of shoot tip,the induction effects were much better and saved time and labour.With the increase in NAA concentration in MS medium,the primary somatic embryogenesis and plant regeneration induction rate in cassava variety GR911 showed initial increase followed by a decrease.The proper concentration of NAA was found to be 60.00 mg/L,which could directly induce primary somatic embryogenesis and buds regeneration.Furthermore,out of 9 cassava varieties,the young stems of 8 cassava varieties viz.,GR911,SC5,SC6,SC7,SC8,SC9,Nanzhi188 and SC205 induced the primary somatic embryogenesis and plant regeneration,but the induction rate for different cassava genotypes was different with the highest in SC8 and the lowest in SC7.
