摘要 目的观察非竞争性NMDA受体拮抗剂地艹卓西平马来酸盐(MK-801)在低剂量时对小鼠焦虑、抑郁以及学习记忆能力的影响。方法将昆明小鼠随机分成对照组和MK-801组,采用高架十字迷宫、强迫游泳实验、悬尾实验、开场实验以及Morris水迷宫等方法,检测药物对小鼠行为的影响。结果与对照组相比,MK-801组抑郁模型中的不动时间减少(P<0.01);焦虑模型中在入开臂次数及停留时间的百分比增加(P<0.01),入臂总次数与总时间也增加;开场实验中运动能力增加(P<0.01);Morris水迷宫中小鼠学习记忆能力下降(P<0.01)。结论低剂量MK-801并未对小鼠产生抗焦虑以及抗抑郁的效果,但可降低学习记忆能力。 Objective To investigate the effect of low dose of N-methyl-D-aspartate(NMDA) receptor antagonist MK-801 on the ethology of anxiety,depression,learning and memory in mice.Methods Male Kunming mice were randomly divided into control group and MK-801 group,and the effect of the drug on the mouse behavior was evaluated by elevated plus maze test,forced swimming test,tail suspension test,open-field test and Morris water maze.Results Compared with the control group,the MK-801 group significantly reduced the immobility duration in the depression model group(P0.01),increase the percentage of open arm entries and open arm duration(P0.01),but also increased the total entries and durations,improved the locomotivity in the open-field test.(P0.01),impaired the ability of learning and memory in the Morris water maze(P0.01).Conclusion Low dose of MK-801 cannot cause anti-anxiety and anti-depressant effects on mice,but can induce impairment of learning and memory.
