九头狮子草水浸膏对2.2.15细胞中对HBsAg、HBeAg的分泌和HBV DNA复制的抑制作用

Inhibitory effect of aqueous extract of Peristrope on secretion of HBsAg and HBeAg and replication of HBV DNA in cultured 2.2.15 cell line

目的:通过观察九头狮子草水浸膏对2.2.15细胞HBsAg、HBeAg的分泌及HBV DNA复制的影响,探讨其在体外的抗乙型肝炎病毒(HBV)作用。方法:将SD大鼠(n=10)随机分为九头狮子草水浸膏组和正常血清组,前者用九头狮子草水浸膏按2.7g.kg-1.d-1剂量灌胃,后者给予生理盐水10ml.kg-1.d-1灌胃,制备不同浓度含药血清及正常血清。2.2.15细胞培养上清液HBsAg、HBeAg测定采用免疫放射分析法,HBV DNA检测采用PCR杂交试验法,测定不同浓度含药血清抗HBV活性情况。结果:含药血清作用4天、8天时,各含药血清组抑制HBsAg、HBeAg分泌的作用优于空白血清细胞对照(指含正常血清)组(P<0.01、P<0.05)。作用4天时,不同浓度含药血清的抗HBV作用差异无显著性意义(P>0.05);作用8天时,不同浓度含药血清抑制HBeAg分泌的作用差异无显著性意义(P>0.05);10%、15%含药血清抑制HBsAg分泌的作用均优于5%含药血清(P<0.05),10%、15%含药血清相比较差异无显著性意义(P>0.05);作用8天时,各含药血清组HBV DNA低于细胞对照组和空白血清细胞对照(指含正常血清)组(P<0.01、P<0.05),5%、10%含药血清组与15%含药血清组相比较差异有显著性意义(P<0.05)。结论:九头狮子草水浸膏的含药血清在体外细胞培养中对2.2.15细胞分泌HBsAg、HBeAg及HBV DNA复制均有较好的抑制作用,10%含药血清的抑制作用优于5%、15%浓度。

Objective：By observing the impact of aqueous extract of Peristrope on the secretion of HBsAg and HBeAg and the replication of HBV DNA in the cultured 2.2.15 cell line,to study the anti-HBV effect of Peristrope in vitro.Methods：SD rats（n=10）were divided into aqueous extract of Peristrope group and normal blood serum group.Rats of the former group were intragastric administerred with aqueous extract of Peristrope with a dose of 2.7 gram per kilogram per day,the latter group were intragastric administerred with normal saline with 10 millilitre per kilogram per day to determined the anti-HBV activities of different concentrations of aqueous extract of Peristrope.The levels of HBsAg and HBeAg in supernatant liquid of cultured 2.2.15 cells were detected by immunoradiometric assay.The levels of HBV DNA were detected by PCR hybridization experiments.Results：Four days and eight days later,the effect inhabiting the secretion of HBsAg and HBeAg in aqueous extract of Peristrope group was better than the effects in the control group and normal serum group（P0.01,P0.05）.Four days later,Anti-HBV effects of extract of Peristrope groups with different concentraions had no significant differences.Eight days later,the effects inhabiting the secretion of HBsAg in extract of Peristrope groups with different concentraions had no significant differences（P0.05）.The effects inhabiting the secretion of HBeAg in 15 percent extract of Peristrope group and in 10 percent extract of Peristrope group were better than the effect in 5 percent extract of Peristrope group（P0.05）and there were no significant difference between 10 percent group and 15 percent group（P0.05）.Four days later,the level of HBV DNA in extract of Peristrope groups with different concentraions were lower than that of the control group and normal serum group（P0.01,P0.05）.The level of HBV DNA in 5 percent extract of Peristrope group and in 10 percent extract of Peristrope group had significant differences with that of 15 percent extract of Peristrope group（P0.05）.Conclusion：Aqueous extract of Peristrope has a significantly inhibitory effect to the secretion of HBsAg and HBeAg and replication of HBV DNA in cultured 2.2.15 cell line in vitro.The inhibitory effect of 10 percent extract of Peristrope is better than that of 5 percent and 15 percent.
