摘要
目的探讨解毒酶基因谷胱甘肽S-转移酶M1(Glutathione S-transferase M1,GSTM1)多态性对黄曲霉毒素B1(aflatoxin B1,AFB1)高污染区广西人群AFB1-DNA加合物水平的影响。方法采用竞争性酶联免疫吸附法检测研究对象(966例)AFB1-DNA加合物水平,而GSTM1基因型分析则通过PCR-RFLP来完成。结果研究结果显示GSTM1缺失型(GSTM1-null)与高AFB1-DNA加合物水平相关,其校正风险值(OR)为2.09(95%可信区间,即95%CI值为1.61-2.71)。结论 AFB1在带有易感基因型GSTM1-null的人群更易诱导产生DNA损伤(AFB1-DNA加合物)。
Objective To investigate the effects of Glutathione S-transferase M1(GSTM1) polymorphism on AFB1-DNA adducts among inhabitants highly exposure to aflatoxin B1(AFB1) in Guangxi province. Methods A competitive enzyme-linked immunosorbent assay was used to measure the AFB1-DNA adducts levels in 966 participants, the GSTM1 genotype analysis completed by using PCR-RFLP.Results The data results indicated that GSTM1 deficiency(GSTM1-null) is positively associated with elevated AFB1-DNA adducts(ajusted odds ratio,OR=2.09,95% confidence interval,95%CI=1.61~2.71).Conclusion AFB1 easily resulted in DNA injuries(AFB1-DNA adducts) among population susceptible to GSTM1-null.
