摘要
利用TUV26菌株所产几丁质酶酶解胶体几丁质制备几丁寡糖粗品,所得产品经凝胶过滤层析将未酶解的几丁质或者聚合度较大的几丁多糖与聚合度在5~10的几丁寡糖分离开。对比研究薄层层析法与荧光标记辅助电泳法分离分析几丁寡糖的效果,结果表明该两种方法都可有效应用于几丁寡糖的检测,其中薄层层析法最佳展层剂为乙酸乙酯∶异丙醇∶水∶氨水=5∶5∶1∶0.3体积比。
Chitinase originated from TUV26 is applied to prepare chitooligosacchrides(COS)through the enzymolysis of colloidal chitin. The resulted products contain unlysis chitin or chito-polysacchrides strain and COS with its drgree of polymerization being from 5 to 10, all of which are separated by gel-filtration chromatography. Two kinds of method for separating and evaluating chito-oligosacchride, thin-layer chromatography and Fluorophore-assisted Carbohydrate Electrophoresis, have been compared for their separation and evaluation effects. The results show that both of them can been utilized in the analysis of COS, and also the best developing reagent is acetidin/ isopropanol/water/ ammonia solution (=5∶5∶1∶0.3 v/v/v/v).
