摘要 目的:探讨蛇六谷乙酸乙酯萃取物A1组分对人胃癌SGC-7901细胞株的增殖抑制作用及其作用机制。方法:用MTT法检测不同浓度A1组分对SGC-7901细胞的增殖抑制作用,光镜学观察A1组分对SGC-7901的细胞形态学变化,RT-PCR法观察A1组分对SGC-7901细胞bcl-2及Bax基因表达的影响。结果:A1组分对SGC-7901细胞增殖抑制作用为剂量依赖性,并具有明显的细胞形态学改变;RT-PCR结果显示,随着A1组分剂量的增加,具有明显的下调bcl-2基因及上调Bax基因水平的作用。结论:A1组分对人胃癌SGC-7901细胞的增殖具有明显的抑制作用,其抑制作用可能与下调bcl-2基因及上调Bax基因水平作用有关。 Objects: To study the effect of the fraction A1 separated from acetic ether extract of Amorphophallus Konja on multiplication of humor stomach cancer SGC-7901 cells and its mechanism.Methods: The effects of fraction A1 on multiplication of humor stomach cancer SGC-7901 cells were observed by MTT reduction assay,the cellular morphologic changes observed by spectroscopy,and the expression of bcl-2 and Bax genes observed by RT-PCR.Results: Fraction A1 showed evident inhibitory action on multiplication of SGC-7901 cells and had a dose-effect relationship,and further exhibited evident cellular morphologic changes.The bcl-2 decreased and Bax increased were also observed with fraction A1 dosage increasing.Conclusion:The fraction A1 separated from acetic ether extract of Amorphophallus Konja has a evident inhibitory action on multiplication of humor stomach cancer SGC-7901 cells,and its mechanism may be related with the bcl-2 decreased and Bax increased.
