摘要
目的:研究蒲黄提取物对氧化低密度脂蛋白(Ox-LDL)诱导人脐静脉内皮细胞(HUVECs)脂质过氧化的影向。方法:体外培养人脐静脉内皮细胞,采用MTT法测定不同浓度蒲黄提取物对内皮细胞有无毒性。根据MTT法的结果,采用蒲黄提取物(10mg/L,50mg/L,100mg/L)预处理细胞24h,再加入100mg/L Ox-LDL造成细胞损伤,检测细胞培养液中乳酸脱氢酶(LDH)的活性、丙二醛(MDA)的含量及超氧化物歧化酶(SOD)的活力。结果:Ox-LDL损伤组LDH的活性、MDA的含量显著升高,SOD的活力显著降低,与正常对照组比较有显著差异(P<0.05);10、50、100mg/L蒲黄提取物可显著降低LDH的活性、MDA的含量,提高SOD的活力。结论:蒲黄提取物可通过抗脂质过氧化,清除自由基起到保护血管内皮细胞的作用。
Objective:To study the effect of pollen typhae extract on lipid peroxidation in human umbilical vein endothelial cells(HUVECs) induced by oxidized low density lipoprotein(Ox-LDL) in vitro.Methods:The human umbilical vein endothelail cells(HUVECs) were cultured in vitro,and different concentration of pollen typhae extract was added to the media for intervention.The influence were detected with MTT assay.According to the MTT,HUVECs were exposed to pollen typhae extract(10mg/L,50mg/L,100mg/L)for 24 h,then treated with 100mg/L Ox-LDL to establish endothelial cell injury model.The activity of Lactate dehydrogenase(LDH),superocide dismulase(SOD) and the content of Malondialdehyde(MDA) in the HUVECs supernatants was determined.Results: Ox-LDL induced the increase of the activity of LDH and MDA,and the decrease of the activity of SOD.The difference was significant as compared with the control group(P〈0.05);Pollen typhae extract(10,50,100mg/L) decreased the the activity of LDH and MDA,and increased the activity of SOD.Conclusion:Pollen typhae extract protects endothelial cells against lipid peroxidation insult by scavenging the free radicals.
