摘要
目的:探讨Fos与磷酸化的转录因子环磷酸腺苷反应元件结合蛋白(cAMP responsive element binding protein,CREB)在正常昆明小鼠前脑的表达及相互关系。方法:成年昆明小鼠,4%多聚甲醛灌注固定、取脑、冰冻切片30μm厚,应用免疫组化法检测Fos和磷酸化CREB(phosphorylated CREB,pCREB)的表达。结果:(1)前脑Fos与pCREB表达较高且一致的脑区主要在前扣带回皮质、顶皮质、梨状前皮质、扣带后回和压部皮质等;(2)二者表达一致但较低的脑区,包括海马CA1、CA3区、隔内侧核、苍白球、梨状内核和杏仁基外侧核等。结论:在生理条件下,Fos在边缘系统的较高表达与边缘系统功能调节有关,pCREB在边缘系统的较高表达与生理性刺激的诱导有关,也与边缘系统的调节有关,二者在前脑的表达分布表明CREB的磷酸化启动了Fos的表达。
Objective:To investigate Fos and cAMP responsive element binding protein(pCREB)basal expressions in the forebrain of Kunming mice and the correlatation between each other.Methods:The adult Kunming mice were sacrificed and fixed with 4% paraformaldehyde.The brains were removed and cut into 30 μm sections.The immunohistochemi-cal staining method was used to detect the expressions of Fos and pCREB.Results:(1)The higher and consistent basal expressions of Fos and pCREB in the forebrain regions were mainly in anterior cingulated cortex(ACC),parietal cortex(Par),prepiriform cortex(PPir),posteriorcingulated cortex(PC)and retrosplenial cortex(RS),etc.(2)The consistent but lower expressions of Fos and pCREB in the following regions:cornu ammonis 1(CA1),cornu ammonis 2(CA3),medial septal nucleus(MS),globus pallidus(GP),endopiriform nucleus(En)and basolateral amygdaloid nucleus(BL),etc.Conclusion:In physiological condition,the higher expressions of Fos were related with the regulation of limbic function.The higher expressions of pCREB were related with the induction of physiological stimuli,and also with the regulation of limbic function.The distribution of Fos and pCREB expressions in the forebrain indicates phosphorylation of CREB could initiate the expressions of Fos.
