半胱氨酰白三烯受体在大鼠分泌性中耳炎黏膜中的表达 被引量:3

摘要 目的探讨半胱氨酰白三烯受体(CysLTR)-1、2在变应性分泌性中耳炎黏膜中表达及其变化。方法建立卵白蛋白激发变态反应中耳炎大鼠模型。实验动物分为阴性对照组Ⅰ、阴性对照组Ⅱ和实验组,采用半定量反转录-聚合酶链反应(RT-PCR)技术分别检测半胱氨酰白三烯受体CysLTR1、CysLTR2 mRNA在中耳黏膜的表达变化。结果 CysLTR1、CysLTR2 mRNA在阴性对照组Ⅰ、Ⅱ的中耳黏膜表达很低,炎症情况下两型受体在实验组中耳黏膜的表达均显著增高,相对值分别为(0.42±0.05)和(0.28±0.06),与阴性对照组Ⅰ、Ⅱ比较差别有统计学意义(P<0.01)。结论 CysLTR1、CysLTR2均在中耳黏膜表达,变应性炎症时表达明显上调。 Objective To determine the expressions of cysteinyl leukotriene receptors,CysLTR1 and CysLTR2,in a rat model of otitis media with effusion induced by ovalbumin(OVA).Methods Eighteen Sprague-Dawley rats were divided into three groups(the experimental group,negative control Ⅰ and Ⅱ).Rats from the experimental group were sensitized to OVA via intraperitoneal injection once a week for two weeks and then challenged transbullarly.Middle ear mucosas were harvested and assayed for CysLTR1 and CysLTR2 mRNA expressions by semi-quantitative reverse transcription-polymerase chain reaction.Results The mRNA expression of CysLTR1 and CysLTR2 was detectable in middle ear mucosas of the negative control groups and significant up-regulation was observed after OVA challenge(P0.01).Conclusion Two types of receptors of cysteinyl leukotriene are present in the middle ear mucosa and can be modulated immunologically.
