摘要
目的探讨姜黄素对大鼠实验性胃溃疡的作用及机制。方法将40只大鼠随机分为4组,即正常对照组、溃疡模型组、姜黄素组、奥美拉唑组。采用水浸-束缚(WRS)方法建立大鼠胃溃疡模型,检测各组胃黏膜溃疡指数(UI)、胃液pH值、壁细胞H+,K+-ATP酶活性和mRNA基因表达,采用SPSS13.0软件进行统计学分析。结果与正常对照组相比,溃疡模型组UI明显升高(P<0.01),胃液pH值明显下降(P<0.01),壁细胞H+,K+-ATP酶活性和mRNA基因表达明显升高(P<0.01);与溃疡模型组相比,姜黄素组UI明显下降(P<0.01),胃液pH值明显上升(P<0.01),壁细胞H+,K+-ATP酶活性和mRNA基因表达明显下降(P<0.01);姜黄素组与奥美拉唑组相比差异无统计学意义(P>0.05)。结论姜黄素可有效抑制胃溃疡模型大鼠胃壁细胞H+,K+-ATP酶活性和mRNA基因表达,减少胃酸分泌,减轻胃黏膜损伤,对胃溃疡有良好的防治作用。
Objective To observe the effect of curcumin on stress-induced gastric ulcer and to explore its mechanism in rats. Methods Forty male SD rats were divided into four groups randomly: control group, gastric ulcer group, curcumin-treated group and omeprazole-treated group. The rat gastric ulcer model was established by water immersion-restraint stress (WRS). The gastric mucosa ulcer index (UI), gastric juice pH, H+,K+-ATPase activity and mRNA expression were measured. The results were analyzed with the SPSS 13.0. Results Compared with control group, the UI, H+, K+-ATPase activity and mRNA expression increased significantly in gastric ulcer group (P 〈 0.01 ), and pH value decreased significantly (P 〈 0.01) in gastric ulcer group. Compared with gastric ulcer group, the UI, H~, K^-ATPase activity and mRNA expression decreased significantly in curcumin-treated group (P 〈 0.01 ), but, the pH of gastric juice increased significantly (P 〈 0.01 ). There were no significant difference of UI, pH, H+, K+- ATPase activity and mRNA expression between curcumin-treated group and omeprazole-treated group (P 〉 0.05). Conclusions Curcumin can effectively inhibit H+, K+-ATPase activity and mRNA expression, reduce gastric acid secretion, and protect gastric mucosal from injury, which suggest that curcumin may have a potential preventive and treatment effect on gastric ulcer.
