摘要
Bir1p是酵母凋亡途径中抑制细胞凋亡的重要蛋白,Survivin是Bir1p的人源同源物,Survivin第34位氨基酸T向A的突变能使其功能逆转。将Survivin及其突变体Survivin(T34A)的基因分别构建到组成型穿梭表达质粒pGAPZA中,整合入毕赤酵母的基因组,分析对酵母细胞凋亡的影响。酵母生长曲线、MTT及流式细胞仪测定数据显示,Survivin和其突变体基因在酵母中的表达分别抑制和促进了酵母凋亡。多样的工艺对工业用酵母的凋亡提出了不同的要求,本研究为工程酵母的理性改造提供了理论依据。
Bir1p plays an important anti-apoptotic role in yeast apoptosis pathway.The mutant of Threonine(34)to Alanine converts Survivin,which is as the homologue in human of Bir1p,from an anti-apoptotic to a pro-apoptotic factor.To study the effect of Survivin and Survivin(T34A)on yeast growth,the both genes were integrated into Pichia pastoris genome with pGAPZA.The growth curve,MTT and Flow Cytometry,were applied to reflect the effect.The result suggested that Survivin and Survivin(T34A)acted as an anti-apoptotic and pro-apoptotic factor in Pichia pastoris death pathway,respectively.Multiple process require various characters of yeast,and the paper could provide the effective approach to reconstruct the engineering yeast.
