摘要 本研究旨在探讨Cu2+浓度梯度、胁迫时间对禾花鲤(Procypris mterus)血液和肝脏的过氧化氢酶(catalase,CAT)活性的影响,为禾花鲤的重金属胁迫研究及水污染生物监测提供理论参考依据,进一步指导禾花鲤的健康养殖。采用可见光法测定了不同Cu2+浓度(0、0.067、0.084、0.101、0.118和0.134 mg.L-1)和胁迫时间(0、1、5和11 d)下禾花鲤CAT活性的变化。结果显示,在试验的各个持续时间段(1、5和11 d),低Cu2+浓度处理组(0.067 mg.L-1)血液及肝脏的CAT活性高于其它各处理组,且活性随着胁迫时间的延长而增加。而中、高Cu2+浓度处理组(0.084、0.101、0.118和0.134 mg.L-1)血液和肝脏中的CAT活性则随着Cu2+浓度的升高和胁迫时间的延长被显著抑制(P<0.05),活性变化在5 d后趋于稳定。试验表明鱼体过氧化氢酶的活性受水体中Cu2+浓度的影响,低Cu2+浓度能诱导鱼体血液和肝脏中的CAT活性增加,但随胁迫的加剧,CAT活性受到抑制。 The aim of this experiment was to examine the effects of copper stress on the catalase(CAT) activity in blood and liver tissue of Carp(Procypris mterus).Carps were treated with 0,0.067,0.084,0.101,0.118 and 0.134 mg·L-1 of Cu2+ for periods of 0 to 11 d under visible light condition.The results showed that highest CAT activities in blood and liver tissue were observed in the lower Cu2+ concentrations treatment(0.067 mg·L-1)and CAT activities of this treatment were increased with extension of exposure time.However,CAT activities in blood and liver tissue in other treatments(0.084,0.101,0.118 and 0.134 mg·L-1 Cu2+ concentrations)were decreased with Cu2+ concentrations and exposure time(P〈0.05)and tended to be stable after 5 days.The experiment suggests that CAT activity can be affected by Cu2+ concentrations in water,and a low dose stimulation followed by a high dose inhibition.
