两种化学表面芯片对SELDI-TOF MS结果的影响

摘要 目的采用标准化芯片操作规程比较弱电离子交换芯片(CM10)、金属离子整合芯片(I MAC)对增强激光解析电离飞行时间质谱(SELDI-TOF MS)检测结果的影响。方法收集1份健康体检者血清,每一个芯片类型都检测8孔。分析质谱峰的数量(S/N>5、S/N>2.5、S/N>10),计算孔间变异系数(CV),通过比较这两个指标来评估两种芯片对血清蛋白组的检测能力。结果在合适的激光能量强度下,S/N>5时,IMAC所测得的质谱峰有247个,CM10所测得的质谱峰有167个,两者的质谱峰平均CV值相近,均在20%左右。结论 IMAC有更好的检测能力。 Objective To compare the influences of CM10 and IMAC chips on the detection results of surface-enhanced laser desorption/ionization time of flight mass spectrometry(SELDI-TOF-MS)by adopting the standardization chip operating protocols.Methods One serum sample taken from one healthy volunteer was applied in 8-hole detection for each chip type.To analyze the total number of detected peaks(S/N〉5,S/N〉2.5,S/N〉10)and to determine the inter-hole variation coefficient by calculating the average CV of peak intensities.The detection ability to serum proteomics was evauated by comparing the two indexes.Results Under proper laser energy intensity,S/N5,IMAC showed 247 detected peaks,CM10 showed 167 detected peaks,the average CV of peak intensities of two chips were similar.Conclusion IMAC has better detecting ability.
