摘要
目的探讨锚蛋白重复和激酶域1(ANKK1)基因多态性与纳洛酮治疗安眠药中毒疗效的影响及临床意义。方法运用聚合酶链反应扩增及单核苷酸多态性的分子生物学技术,对应用纳洛酮治疗安眠药中毒患者78例和78例正常人ANKK1基因的rs4938015、rs7118900、rs2734849、rs1800497多态性分型,并对ANKK1基因多态性与纳洛酮治疗安眠药中毒之间关系进行比较分析。结果在纳洛酮治疗安眠药中毒后2h患者GCS评分的数值比较,rs2734849位点差异有统计学意义(F=5.89,P〈0.05),且rs2734849位点中以纯合子T/T评分数值更高;各SNP位点与疗效的关系比较,rs2734849位点差异有统计学意义(X^2=8.72,P〈0.05),其余各位点比较差异无统计学意义。结论ANKK1基因与纳洛酮治疗安眠药中毒的疗效存在明显的相关性。
Objective To study the clinical effect of naloxone treatment on sleeping pills poisoning and polymorphism of Ankyrin repeat and kinase domain containing 1 ( ANKK1 ) gene. Methods Observed in a sample of 78 parents whose poisoning of sleeping pills were treated by naloxone. The Glasgow coma scale would be assessed in 0 and 2 hours on the cases, while assessing on the advantages and the disadvantages of naloxone efficacy. The other sample of 78 normal people would be control group. The polymorphism of rs4938015 ,rs7118900 ,rs2734849,rs1800497 of ankyrin repeat and kinase domain containing 1 gene was detected with PCR and SNP typing in two samples. The relationship between the ankyrin repeat and kinase domain 1 gene polymorphisms and treatment effect was analyzed. Results The rs2734849 allele is significant connected with naloxone treatment on sleeping pills poisoning through the Glasgow coma scale ( F = 5.89, P 〈 0. 05 ) and the efficacy ( X2 = 8.72, P 〈 O. 05 ). The homozygotes T/T had better efficacy than T/A and T/T. The rs4938015, rs7118900 and rs1800497 allele were not significant with naloxone treatment. Conclusion It shows an association between naloxone treatment on sleeping pills poisoning and the polymorphism at nucleotide of Ankyrin repeat and kinase domain containing 1 gene in Chinese.
