摘要
目的在蛋白质水平探讨3-硝基丙酸(3-NAP)预处理对缺血-再灌注损伤(IRI)大鼠的脑保护作用的可能机制。方法采用大脑中动脉线栓法(MCAO)制备脑缺血再灌注模型,对3-NPA预处理的大鼠(预处理组)与单纯IRI组大鼠(IRI组)应用荧光差异双向凝胶(2D-DIGE)电泳、基质辅助激光解析/电离-飞行时间质谱分析(MALDI-TOF MS)及数据库检索鉴定出差异表达的神经细胞骨架蛋白。结果预处理组与IRI组相比有52种蛋白表达量具有显著性差异,其中鉴定出15种主要的差异蛋白质,神经细胞骨架相关蛋白有7种。结论一些神经细胞骨架相关蛋白可能对IRI后的脑组织具有保护作用。
Objective A preliminary study of the potential mechanism of protection of 3-nitropropionic acid pretreatment on ischemia-reperfusion injury of rat brain at the protein level.Methods The rat model of IRI was established by middle cerebral artery occlusion(MCAO),and differentially expressed nerve cytoskeletal proteins were identified between 3-NPA pretreated rats(preconditioning group)and simply IRI group(IRI group)by two-dimensional difference gel electrophoresis(2D DIGE)technique,combined with matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF MS).Results 52 proteins were expressed differentially of the pretreatment group compared to the control group 72h after surgery,15 main expressed proteins of which was identified,7 ones of which were nerve cytoskeletal associated protein.Conclusion Some nerve cytoskeletal associated proteins have protective effects against nerve ischemia reperfusion injury.They may be the target for the neuro-protective effect of 3-NPA pretreatment.
