摘要
应用PK15细胞从山东省某猪场分离到1株病毒,通过免疫过氧化物酶单层试验和全基因组序列分析,证明该分离株为猪圆环病毒2型(PCV-2,命名为SD/2008株),基因组全长1767bp,为PCV-2b基因型;感染PK15细胞后96h病毒含量最高,培养液含病毒基因组1.72×108copies/μL,连续培养120h不出现细胞病变;连传5代,病毒滴度稳定,维持在106TCID50/0.1mL左右;传代PK15细胞时同步接种病毒后18h,用300mmol/L的D-氨基葡萄糖处理细胞,病毒滴度可以提高5.62倍。
A strain of porcine circovirus type 2,named PCV-2 SD/2008 strain,was isolated with PK15 cell from a pig farm in Shandong province and identified by immunoperoxidase monolayer assay(IPMA) and complete genomic sequence analysis.Its genome contained 1 767 bp and belonged PCV-2b genotype;after 96 h innoculation,the content of virus in PK15 cell reached the highest,1.72×108 viral DNA copies per microlitre;and no cytopathogenic effect appeared within 120 h cultivation;the virus titer stayed at the level of about 106 tissue culture infective dose 50 (TCID50/0.1 mL) within passage 2 to 5 and increased by 5.62 times when the PK15 cells were treated with 300 mmol/L of D-glucosamine at the 18th hour after synchronized inoculation.
