摘要
通过比较蛋白质组学分析M5生存表型的改变所引起的蛋白图谱与强毒株的差异之处,根据这些差异表达蛋白的LC/MS-MS鉴定结果来分析疫苗株的致弱机制进而为寻找新的毒力相关分子、鉴别诊断分子以及揭示布鲁菌胞内寄生机制提供帮助。本研究利用比较蛋白质组学技术,对布鲁菌强毒株16 M和弱毒株M5的外膜蛋白进行了差异比较分析,结果共发现33个差异蛋白点,代表了26个开放阅读框,这些蛋白涉及了蛋白质的生物合成(5/26)、氨基酸合成(3/26),脂肪酸代谢(2/26)、能量代谢(5/26),以及细胞被膜生物合成(4/26)和一些调节系统(4/26)等多个生物过程。这些发现为研究外膜蛋白在布鲁菌毒力,胞内寄生等过程中发挥的重要作用提供了新依据。
A comparative analysis of the outer membrane proteins of Brucella melitensis 16M and M5 was performed using two-dimensional gel electrophoresis and the proteins of interest were identified using LC-MS/MS,which would be helpful for us to find new virulent factors,differential diagnosis molecules and reveal the intracellular survival mechanism for Burcella spp in host cell.In this study,a difference map was created for B.melitensis 16M and M5 containing 33 entries proteins,which represent 26 discrete open reading frames(ORFs) and are closely correlated with a number of bioprocesses,such as protein synthesis,amino-acid-biosynthesis,cell-wallbiogenesis,energy-metabolism,regulatory functions,fatty acid and phospholipid metabolism.These findings would provide insight for the roles of Brucella Omps about bacterial virulence and intracellular survival.
