摘要
目的:建立射干中芒果苷的含量测定方法;测定不同产地射干中芒果苷的含量。方法:采用RP-HPLC法,外标法定量,Kromail C18柱分离,流动相:乙腈-0.2%磷酸水溶液(15∶85),检测波长257 nm,柱温为25℃,流速1mL.min-1。从湖北、安国、北京及沈阳分别采集射干供试品;对湖北及安国射干根茎与须根中芒果苷的含量进行比较。结果:所建立的RP-HPLC含量测定方法线性关系良好(r>0.9990),方法平均回收率98.65%,RSD=1.62%(n=5)。结论:所建立的RP-HPLC含测方法简单,快速,专属性强好。不同产地射干中芒果苷的含量有差异,须根中芒果苷含量显著高于根茎中。
Objective:To establish an HPLC method for determination of Mangiferin in Belamcanda.By thisnew method,Mangiferin in Belamcand was determinated.Methods:The assay was performed on a Kromail-C18 column(200mm×4.6mm,5μm),eluted with a mobile phase consisted of acetonitrile and 0.2% phosphate and gradient elution at a flow rate of 1mL/min.The detection wavelength was 257nm.Results:Excellent chromatographic separation was obtained for mangiferin with good linearity(r0.9990),the average recovery were 98.65%,RSD=1.62%(n=5).Conclusion:The method is simple,rapid and specific.The contentof Mangiferinin was statistically different due to different areas or parts.The content of mangiferin in beard.were higher than in rhizome.
