眼针疗法对IBS模型大鼠结肠组织色氨酸羟化酶1表达的影响 被引量:2

Effects of Ocular Acupuncture Therapy on Colonic Tryptophan Hydroxylase 1 in the Rat with Irritable Bowel Syndrome

摘要 目的:研究肠易激综合征(Irritable bowel syndrome,IBS)模型大鼠远端结肠组织色氨酸羟化酶1(Tryptophan hydroxylase1,TPH1)的表达以及眼针干预的影响。方法:实验动物分成正常对照组、IBS模型组、眼针组,采用束缚刺激与应激刺激相结合的方法建立IBS动物模型并进行眼针干预。各组采用Western-Blot法、RT-PCR法检测远端结肠组织中TPH1的蛋白和mRNA表达。结果:与正常对照组比较,IBS模型组TPH1的蛋白和mRNA表达均上调;与IBS模型组比较,眼针组TPH1表达下调明显。结论:IBS模型大鼠结肠组织TPH1表达上调,提示5-HT合成水平提高,可能是结肠组织5-HT水平升高的重要原因。眼针干预可改善IBS大鼠TPH1表达至正常水平。 Objective:To investigate alteration of tryptophan hydroxylase1(TPH1) of distal colonic mucosa in the rat with irritable bowel syndrome(IBS) and effects of ocular acupuncture therapy(OAT).Methods:Animals were classified into blank control、IBS model and OAT groups.Rat model of IBS was established by method of stress and combining bound,and OAT was given.The mRNA and protein content of TPH1 at distal colonic mucosa of each group was examined by Western-Blot and RT-PCR.Results:The mRNA and protein expression of TPH1 in IBS model increased remarkably compared with those in control.The expression of TPH1 in OAT decreased remarkably compared with those in IBS model.Conclusion:The synthesized excessive 5-HT function of colonic TPH1 may play an important role in the high 5-HT of IBS,OAT can correct TPH1 function effectively.
