双水相体系在糖化酶提取纯化中的应用研究 被引量:2

摘要 本文对PEG/KH2PO4、PEG/(NH4)2SO4和PEG/MgSO4三种体系进行了研究,通过制作相图,分析得出PEG/(NH4)2SO4双水相体系是萃取糖化酶最合适的体系。研究了PEG分子量、PEG溶液浓度、(NH4)2SO4溶液浓度对糖化酶分配系数、回收率的影响,确定了萃取糖化酶的最佳条件,即PEG分子量为20000、PEG溶液浓度为28%、(NH4)2SO4溶液浓度为20%时,分配系数为0.15,糖化酶回收率最高为96.1%。 PEG/KH2PO4,PEG/(NH4)2SO4 and PEG/MgSO4 aqueous two-phase systems were studied.Result indicated that PEG/(NH4)2SO4 aqueous two-phase system was best.Then the glucoamylase was extracted and purified by PEG/(NH4)2SO4 aqueous two-phase system.The effects of the molecular of PEG,the concentration of PEG and PEG/(NH4)2SO4 solution on distribution coefficient and recycling rate were studied.The optimum conditions for glucoamylase extraction were determined.When the molecular of PEG was 20000,the concentration of PEG20000 solution was 28% and that of(NH4)2SO4 solution was 20%,the smallest distribution coefficient and highest recycling rate were obtained as 0.15 and 96.1%,respectively.
