摘要
目的探讨Fas/FasL系统对LPS诱导肺泡Ⅱ型上皮细胞分泌炎症因子(MCP-1、TNF-α)的影响。方法原代培养肺泡Ⅱ型上皮细胞,随机分为四组(每组平行6孔):对照组、LPS组、LPS+FasL组、FasL组,加入LPS、FasL调节终浓度为LPS:10μg/ml,FasL:5 ng/ml。培养48 h后,ELISA检测MCP-1、TNF-α浓度。结果与对照组比较,LPS组、FasL+LPS组、FasL组TNF-α与MCP-1浓度增加(P<0.01);与LPS组比较,FasL+LPS组TNF-α与MCP-1浓度增加(P<0.05)。结论 Fas/FasL使LPS诱导的肺泡Ⅱ型上皮细胞MCP-1与TNF-α分泌增加。
Objective To study the effect of Fas/FasL on secretion of MCP-1 and TNF-α from rat alveolar type Ⅱ epithelial cells induced by lipopolysaccharide(LPS).Methods Rat alveolar type Ⅱ epithelial cells in vitro which were isolated,purified and incubated were randomly divided into four groups: control group,LPS group,LPS+FasL group,and FasL group.LPS and FasL were put into DMEM.The final concentrations of LPS and FasL were 10 ug/ml and 5 ng/ml,respectively.After incubation for 48 hours,the concentrations of MCP-1 and TNF-α were measured by ELISA.Results As compared with control group,the concentrations of MCP-1 and TNF-α of rat alveolar type Ⅱ epithelial cells in vitro were significantly increased in LPS group,FasL+LPS group and FasL group(P〈0.01).As compared with LPS group,the concentrations of MCP-1 and TNF-α were significantly increased in FasL+LPS group(P〈0.05).Conclusions Fas/FasL can increase secretion of MCP-1 and TNF-α from rat alveolar type Ⅱ epithelial cells induced by LPS.
