小白菊内酯对TGF-β1诱导的大鼠肺成纤维细胞转分化的影响 被引量:1

摘要 目的探讨小白菊内酯(PNT)对转化生长因子-β1(TGF-β1)诱导的大鼠肺成纤维细胞转分化的影响。方法选择12周龄健康SD大鼠12只,组织块法体外培养大鼠肺成纤维细胞,免疫细胞化学染色,观察不同浓度PNT对TGF-β1诱导的肺成纤维细胞表达α-平滑肌肌动蛋白(α-SMA)的影响。结果 TGF-β1诱导肺成纤维细胞转分化为肌成纤维细胞,表达α-SMA增强(P<0.05),20μmol/L的PNT部分抑制TGF-β1的诱导作用(P<0.05);50μmol/L的PNT可完全抑制TGF-β1的诱导作用。结论 PNT有抑制TGF-β1诱导肺成纤维细胞转分化的作用。 Purpose To investigate the influence of Parthenolide on the rat′s lung fibroblast trans-differentiation which was induced by the transforming growth factor-β1(TGF-β1).Methods Were choosen 12 healthy SD rats of 12 weeks of age and were be cultured rat lung fibroblasts by the explanted method.Then there would be chemical staining of the immune cells to observe the influences of PNT with different densities on theα-SMA which was induced by TGF-β1.Results The TGF-β1 induced the pulmonary fibroblast to be transformed into myofibroblasts and increased the expression of α-SMA(P0.05).20 μmol/L of PNT partially inhibited the induction of TGF-β1(P0.05),50 μmol/L of PNT completely inhibited the induction of TGF-β1.Conclusion PNT could inhibit the transdifferentiation of TGF-β1 induced pulmonary fibroblast.
