Roux-en-Y胃旁路术对GK大鼠血糖水平影响的研究

Research into Hypoglycemic Effects of Roux-en-Y Gastric Bypass on Goto-Kakizaki Rats

目的:研究保留胃容积Roux-en-Y胃旁路术(RYGB)对GK大鼠血糖水平的影响。方法:将30只3月龄雄性GK大鼠随机分为空白对照组、假手术组、RYGB组,观察并记录各组术前术后体重、饮水量,用口服糖耐量试验(OGTT)测量手术前、后相同时间位点空腹和180min血糖值,并观察糖化血红蛋白值的变化。结果:各组GK大鼠实验后各时间段体重较实验开始前均有明显变化(P<0.05);RYGB术后GK大鼠的饮水量较术前空白组及假手术组明显下降(P<0.05);RYGB术后所有GK大鼠的空腹血糖均较术前逐渐降低(P<0.05),术后180min血糖均较术前空白组及假手术组降低(P<0.05);RYGB术后GK大鼠的HbA1c较术前空白组及假手术组逐渐下降(P<0.05)。结论:提示保留胃容积RYGB对GK大鼠在不影响体重增加的同时从行为学,生物学指标上都有明显降低血糖的作用。

Objective： To study hypoglycemic effects of Roux-en-Y gastric bypass surgery on Goto-Kakizaki rats.Methods： Thirty three-month old male GK rats were divided into three groups： negative control group,sham-operation group and RYGB group.Their body weight and the amount of water they drank before and after the operation were respectively observed and recorded.Oral glucose tolerance test（OGTT）measuring blood glucose levels in a fasting state and 180 minutes after oral glucose was performed before and after the operation to measure the levels of glucose in GK rats plasma,and the levels of hemoglobin（Hb） in blood were also detected before and after the operation.Results： There was a significant difference in body weight before and after the experiment in all three groups（P0.05）.After RYGB surgery,GK rats＇ amount of drinking water was lower than preoperative levels and that of the other two groups（P0.05）.The plasma levels of FPG decreased gradually after RYGB operation in all GK rats（P0.05）.And the 180 minutes level of glucose in RYGB group were lower than preoperative levels and that of the other two groups（P0.05）After RYGB operation,the levels of HbA1c in RYGB group seemed to be lower than preoperative levels and that of the other two groups（P 0.05）.Conclusion： From ethological and biological indexes,our data indicated that Roux-en-Y gastric bypass could reduce blood glucose levels significantly without affecting weight gaining.
