摘要
目的:考察和厚朴酚在HBSS溶液中的溶解性和稳定性,为和厚朴酚在体内的吸收与代谢研究提供依据。方法:采用RP-HPLC法,分别考察了pH、温度、光照对和厚朴酚溶解性和稳定性的影响。结果:和厚朴酚在甲醇的标准曲线为Y=4 764.5X-10 955(r=0.999),在4~200μmol·L-1与峰面积呈良好的线性关系,检测限为0.2μmol·L-1,精密度和稳定性符合要求。和厚朴酚在含有1%甲醇和0.1%的吐温-80的HBSS中的溶解性较好,和厚朴酚在酸性中的稳定性优于碱性环境,在pH 7.4的HBSS中的稳定性比较理想,光照对和厚朴酚的稳定性没有影响。结论:和厚朴酚在pH 7.4的HBSS中的溶解性和稳定性比较理想,当其达到溶解平衡之后于-20℃储存。
Objective:To investigate the solubility and stability of honokiol in HBSS,Establish the basis of the drug metabolism research.Method:The concentration of honokiol was determined by RP-HPLC method,pH,temperatures and light were investigated on the solubility and stability of honokiol.Result:The linear calibration curve of Honokiol in methanol in the concentration range of 4-200 μmol·L^-1 with a detection limit of 0.2 μmol /L,The equation was calculated to be Y = 4 764.5X-10 955(r=0.999),The precision and stability accorded with the requirements of methodology.The solubility of honokiol is good in HBSS containing 1% methanol and 0.1% Tween-80,the stability of honokiol in HBSS of acidic is more better than HBSS of alkaline,.light on the stability of honokiol has no influence.Conclusion:Tthe solubility and stability of honokiol in HBSS of pH 7.4 is rather ideal,when it reached the dissolution equilibrium it should be storied at-20 ℃.
