摘要
利用RACE技术,根据已有的三化螟转录组数据,设计特异引物对三化螟的蜕皮激素受体(EcR)和超气门蛋白(USP)基因进行克隆,获得了三化螟EcR和USP基因的全长序列。EcR基因全长2857bp,氨基酸序列包括545个氨基酸。USP基因全长共2082bp,氨基酸序列包括408个氨基酸。序列比对后发现,三化螟EcR和USP基因的氨基酸序列与已报道的鳞翅目昆虫EcR和USP基因的氨基酸序列具有很高的同源性,其中与二化螟的相似性分别达到94%和97%。序列分析表明,三化螟EcR和USP序列具有昆虫EcR和USP基因的特征结构:DNA结合域和2个锌指结构。
In this paper,specific primers were first designed based on transcriptome data to verify gene fragments of Tryporyza incertulas. The full-length EcR and USP gene were cloned by RACE techniques. The full-length EcR cDNA was 2 857 bp,with an open reading frame encoding 545 amino acids. And the full-length USP cDNA was 2 082 bp,with an open reading frame encoding 408 amino acids. Blast analysis demonstrated that EcR and USP gene had more than 80% similarity in amino acid sequence with other members of EcR and USP from lepidopteran. The highest similarity was found with the EcR and USP of Chilo suppressalis,up to 94% and 97% ,respectively. Sequence analysis indicated that the EcR and USP of T. inlertalas contained also many common characteristic structures of the EcR and USP family,including DNA binding domain and two zinc fingers in conserved coding region.
