拟南芥抗灰霉病相关基因的克隆与原核表达

摘要 为了从拟南芥中获得抗灰霉病相关基因以及进行基因分析,本试验利用RACE技术获得了该基因的cD-NA全长(1190 bp),并用生物信息学方法和Southern Blotting技术明确了该基因是编码372个氨基酸的转录调节/结合蛋白的AT5G67480基因,以单拷贝形式存在于拟南芥基因组中。编码的蛋白含有BTB/POZ结构域,体外诱导表达的蛋白对灰霉病菌的生长有一定的抑制作用。 Full-length cDNA sequence of resistant-related gene of Arabidopsis against Botrytis cinerea was obtained by running RACE.Result of homology indicated that gene identified as AT5G67480 gene,which encoded binding protein or transcription regulator,containing 372 amino acids.AT5G67480 gene is a single copy in Arabidopsis genomic DNA by Southern Blotting analysis.The protein encodes by AT5G67480 gene contains BTB/POZ domain of TAZ zinc protein family.In most cases,protein containing BTB/POZ domain is related to resistance to pathogens,and such protein is resistant to B.cinerea by expression of AT5G67480 gene in E.coli and in vitro inhibition on B.cinerea.
