比色法测定板栗花中总黄酮的含量 被引量:7

摘要 目的:探索板栗花中总黄酮醇提取工艺的最佳条件,并建立板栗花中总黄酮的含量测定方法。方法:以1%AlCl3甲醇溶液为显色剂,山柰酚-3-O-[6″-O-反式-对-香豆酰基]-β-D-吡喃葡萄糖苷为对照品,采用比色法测定。结果:山柰酚-3-O-[6″-O-反式-对-香豆酰基]-β-D-吡喃葡萄糖苷检测浓度在0.008~0.048mg.mL-1与吸光度呈良好的线性关系,线性回归方程为A=19.089C+0.0263(r=0.9998),平均回收率为96.54%,RSD=0.63%(n=6),用乙醇提取法提取的板栗花中总黄酮含量达6.92mg·g-1。结论:本方法简便、快速、准确、重现性好,可用于板栗花中总黄酮的含量测定。 Objective: To explore the optimal method of extracting total flavones and establish the method for the content determination of total flavonoids in the flowers of Chinese chestnut. Methods: Colorimetry analysis was employed 1% AICI3 worked as colour developing reagent, and kaempferol-3-O-[6″-O-( E)-p-coumaroyl]-β-Dglucopyranoside was used as the reference substance. Results: A good linear relationshi Pwas achieved over the concentration range of 0. 008 -0. 048 mg.mL^-1 for kaempferol-3-O-[ 6″-0-(E)-p-coumaroyl ]-β-D-glucopyranoside with an average recovery of 96.54 %, RSD = 0. 63 % ( n = 6). The equation of linear regression was A = 19. 089 C + 0. 026 3 ( r = 0. 999 8). The total flavones in the flowers of chinese chestnut by ethanol extracting could achieve 6. 92 mg.g^-1 . Conclusion: The method was accurate, rapid, accurate and reproducible, and can be used to determine the total flavones in the flowers of Chinese chestnut.
