摘要
目的:观察氧比修饰低密度脂蛋白(OX-LDL)对血管内皮细胞的损伤作用。方法:在人脐静脉内皮细胞体外培养的基础上,通过采用3H胸腺嘧啶校苷(3H-TdR)掺入DNA技术和硫代巴比妥酸比色法,分别分析了OX-LDL对内皮细胞DNA合成及脂质过氧化物的代谢产物丙二醛(MDA)含量的影响。结果:当内皮细胞暴露于OX-LDL(50~200μg/ml)24小时后,其3H-TdR掺入量明显减少,但MDA含量显著增加。结论:OX-LDL能够明显抑制内皮细胞DNA合成,并显著增加内皮细胞的脂质过氧化物含量。
Objective To study the injury effect and mechanism of Oxidized low density lipoprotein (OX-LDL) on vascular endothelial cells. Methods On the basis of the establishment of culture method for human umbilical vein erdothelial cell (HUVECs), the effects of OX-LDL on DNA synthesis of cultured HUVECs were observed by 3H-thymidine (H-TdR ) incorProation. Moreover, we also measured concentration of malondialde-hyde (MDA) of HUVECs by thiobituric acid reaction. Results The smaples of HUVECs were subjected to 50, 100, 200pg/ml OX-LDL for 24 hour. 3H-TdR incorporation in HUEVCs DNA was decreased significantly, and the content of MDA in HUVECs was increased markedlly. Conclusions OX-LDL could inhibit DNA synthesis of HUVECs, which might be related to lipid Peroxidative injury.
