脑通胶囊对血管性痴呆大鼠海马CA1区突触可塑性的影响 被引量:3

摘要 目的:观察脑通胶囊对血管性痴呆模型大鼠学习、记忆以及海马CA1区突触可塑性的影响。方法:采用改良的四血管法制备VD模型,Morris水迷宫测定大鼠学习、记忆能力,电镜观察海马CA1区突触超微结构的变化。结果:与模型组比较,脑通胶囊大、中剂量组逃避潜伏期缩短,穿越原平台次数增多(P〈0.05-P〈0.01)。模型组突触前、后膜模糊,突触间隙变窄,突触后致密带密度低,突触活性带较短,突触囊泡不清。假手术组突触结构形态正常。大剂量组、中剂量组突触结构形态接近于假手术组。结论:脑通胶囊可减轻海马CA1区突触损伤,从而改善VD大鼠学习、记忆能力。 Objective:To observe the effect of Naotong Capsule on the learning and memory abilities,and the synaptic plasticity in CA1 region of the hippocampus on vascular dementia(VD) rats.Methods:The vascular dementia models were established by modified Pulsinelli 4 vascular occlusions method(4VO).Morris water maze was used to test learning and memory abilities of rats.And the ultrastructure of synapses in CA1 region of the hippocampus was observed by transmission electron microscope.Results:Compare to the model group,the escape time was shortened and times of spanning the previous platform increased in Naotong Capsule large-dose and middle-dose treatment group(P0.05~P0.01).In model group,both presynaptic and postsynaptic membranes were not clear.Synaptic gaps become narrow.The length of synaptic interface shortened,the thickness of postsynaptic density decreased,and the synaptic vesicles appeared darkly or indistinctly.In sham-operation control group,the ultrastructure of synapse was normal.Naotong Capsule large-dose and middle-dose treatment group showed the similar situation as sham-operation control group.Conclusion:Naotong Capsule can improve learning and memory abilities of VD rats by decreasing synapses injury in CA1 region of the hippocampus.
