用两个籼稻品种H359和Acc8558杂交衍生的重组自交系(RI,F7代,131个系)群体,构建了一个密度较高、分子标记分布较均匀的分子连锁图。该图全距1380.1cM,包括147个RFLP和78个AFLP标记,标记间的平均距离为6.1cM。在AFLP分析中,采用5个PstI/MseI的引物组合,总共产生了78个AFLP标记,分布在所有的12条染色体上,平均每个引物组合产生15.6个AFLP标记,表明AFLP是一种高效的分子标记,可与RFLP标记结合。
A linkage map of rice was constructed using an indica×indica(H359×Acc8558) derived population of 131 recombinant inbred(RI) lines(F 7). The map covers 1380.1 cM, consisting of 147 RFLP and 78 AFLP markers with an average distance of 6.1 cM between adjacent loci. The linkage map corresponds well to the RGP map(Kurata et al., 1994) based on F 2 population derived from an inter subspecies cross. Most of the markers were mapped to the same chromosomes and aligned with the same order. Five PstI/MseI primer combinations were used to generate AFLP in the RI population. A total of 78 polymorphic bands were mapped onto the linkage map and distributed over all the 12 chromosomes, demonstrating that AFLP is highly efficient as a type of molecular marker, which can be combined with RFLP marker for the construction of high density linkage maps.
